Understanding Vismodegib and Sonidegib: For Patients

Vismodegib and sonidegib are medications called sonic hedgehog inhibitors, used to treat advanced basal cell carcinoma (BCC) when surgery or radiation are not effective or appropriate. These drugs work by blocking the overactive “hedgehog signaling pathway,” which drives uncontrolled growth of cancer cells in BCC. Clinical studies show they can shrink tumors and, in some cases, clear them entirely. Common side effects include muscle cramps, hair loss, changes in taste, fatigue, and digestive issues, though these usually improve when treatment stops. Doctors may recommend these treatments for patients with advanced, recurrent, or difficult-to-remove BCC, with ongoing follow-up needed to monitor effectiveness and manage side effects.
